The IDT RealTime PCR design tool (found under the SciTools>PCR tab of the IDT website)을 이용해서 Tm을 계산할 때는 realtime PCR 버퍼 조건을 32 mM Na1+, 3 mM Mg2+, and 0.8 mM dNTPs으로 놓고 합니다.
그러나 spec sheet에서는 default conditions( 50 mM Na1+, and does not take into account Mg2+ and dNTP concentrations)에서 계산된 Tm 값을 제공하기 때문에 다른 Tm이 표기됩니다.
그러므로 디자인툴에 의해서 계산된 Tm은 spec sheet 에 나온 값보다 일반적으로 5-8°C 높게 보여집니다.
Real-time PCR 실험에 필요한 Tm 계산은 RealTime PCR design tool 또는 OligoAnlayzer tool 을 이용하되 OligoAnlayzer tool 에서 parameter를 32 mM Na1+, 3mM Mg2+, and 0.2 mM dNTPs 입력 하고 계산하면 됩니다.
