It will be useful in situations where customers are reporting a varying concentration of +/- 20% from the reported value, or if there are concerns regarding oligo purity based upon an absorbance measurement.  It is important to be clear that this is not a problem with the NanoDrop instrument, but rather is related to certain quantification methods having the potential to limit the precision of the measurement.

Technical note regarding oligo quatification using NanoDrop (or similar spectrometer).

 Key points to note:


1. Use oligo-specific conversion factor (i.e. extinction coefficient) instead of general ssDNA conversion factor (i.e. 33ug/OD260) when calculating oligo concentration.
2. Traditional purity ratios (A260/280 and A260/230) are not useful in accessing the purity.
3. Ensure measurements are taken with 1mm (more accurate) or 0.2mm pathlength. Dilute oligo to obtain OD260 of <62.5 or  preferably <12.5 (more accurate).
4. Take duplicate or triplicate measurements.
