The epigenome is regulated by three families of genes: proteins add, bind to, or remove DNA and histone modifications 1,2.  These are commonly grouped as 'Writers,' 'Readers,' or 'Erasers,' respectively.  The shRNA-mir epigenetics focused sets mirror these gene families and conveniently provide shRNA targeting epigenetics-related genes arrayed (in 96 well plates) or pooled for screening.
 
Based on the shERWOOD shRNA design algorithm, every shRNA is expected to knockdown the targeted gene by > 70%.  This consistency increases confidence in results  providing a distinct advantage over other shRNA libraries. Click here to learn more about the shERWOOD algorithm.

 Viral particle format includes:
  • Pool viral particles: 50 µl  at > 10^8 TU/ml
  • Control viral particles: 25 µl  at > 10^8 TU/ml

  • Pool plasmid DNA: 50 µg
  • Control plasmid DNA: 25 µg
 
Screening capacity:
  • Libraries provided as viral particles include enough virus to perform a screen at 1000-fold representation with 3 biological replicates in a cell line with a transduction efficiency similar to HEK293T.
  • Libraries provided as DNA included enough plasmid for more than 3 packaging reactions.  Total viral out will depend on packaging efficiency, but is expected to provide enough viral particles to perform a screen multiple cell lines at 1000-fold representation with 3 biological replicates.
Screening capacity will be impacted by the transduction efficiency of the target cell line.  Please refer to the manual for the necessary calculations. 
 
Arrayed focused sets: 
The complete epigenetics-focused shRNA sets includes genes curated by Dr. Greg Hannon and colleagues at Cold Spring Harbor Laboratory and review literature.  These can be further broken down by activity or functional domain.  Smaller arrayed focused sets are also available targeting genes based on a specific activity or functional domain. See table below. (All focused sets are available in pooled format upon request.)
