Description

Ribonuclease inhibitor effectively inhibits ribonuclease A type(ribonuclease A, B and C) enzyme activity by binding non-covalently to RNase molecules at a 1:1 ratio. It does not inhibit RNA polymerase, DNA polymerase, RNase H, or RNase T1, and is therefore ideal for use in RT-PCR reactions, in vitro tranlscription, or DNA library construction. Mbiotech RNase Inhibitor is a highly purified, nuclease-free preparation. Each lot is rigorously tested and is guaranteed free of non-specific enodnuclease, exonuclease, and RNase activity.

Unit definition

One unit is defined as the amount of ribonuclease inhibitor required to inhibit by 50% activity of 5ng of ribonuclease A. Activity is measured by the inhibition of hydrolysis of cytidine 2¡¯3¡¯-cyclic monophosphate by ribonuclease A, where 200 units of ribonuclease inhibitor produces 50% inhibition of 1ug of ribonuclease A.
 

Standard unit assay conditions

Activity is measured by the inhibition of hydrolysis of cytidine 2',3'-cyclic monophosphate by ribonuclease A, where 200 units of ribonuclease inhibitor produces 50% inhibition of 1¥ìg of ribonuclease A.

Concentration

40 units/¥ìl.

Strorage condition

Avoid multiple freeze/thaw cycles and frequent temperature changes. These fluctuations can greatly affect the product stability.

-Long term(Infrequent use:1-2 times/month):-70¡ÆC.

-Short term(Daily/Weekly use):-20¡ÆC
If stored for long periods at -20¡ÆC or opened frequently during storage, addition of DTT to 5mM final concentration into the reaction mixture is recommended to maintain optimal activity.
Note: Ribonuclease inhibitor is active over a broad pH range but requires a minimum of 1mM DTT to maintain activity.
