ApplicationsUltra-high fidelity for subsequent cloning Blunt-end cloningDescriptionACCUZYME™ is a thermostable DNA polymerase possessing 5´-3´ and 3´-5´ proofreading exonuclease activities. ACCUZYME provides high-fidelity with an error-rate of 3.0 x 10-6, even with demanding applications (fig. 1), resulting in blunt-ended amplicons of up to 5kb in length.
ACCUZYME is supplied with 10x Reaction Buffer containing MgSO4, which provides optimal final reaction conditions (1.5mM Mg2+) for most experiments. In order to allow optimization of reaction conditions, additional MgCl2 is provided.
The specificity and performance of ACCUZYME can be further improved with the use of 2x PolyMate Additive, which is designed for GC or AT-rich DNA, "dirty" templates or sequences with a high level of secondary structure.
The error rate of ACCUZYME DNA Polymerase (3.0 x 10-6) was determined using the rpsL fidelity assay (Lackovich et al., 2001; Fujii et al., 1999). NotesACCUZYME is a trademark of Bioline. Concentration2.5u/μl

Storage & StabilityAll components should be stored at -20°C upon receipt for optimum stability. Repeated freeze/thaw cycles should be avoided.
When stored under the recommended conditions and handled correctly, full activity of the reagents is retained until the expiry date indicated on the outer box label.
